    Thickness of skin layers as a prognostic marker for non-healing wounds

    Nonhealing wounds represent a major clinical challenge, often complicated by infection, delayed recovery, and decreased quality of life. This experimental study investigated the role of skin layer thickness as a prognostic marker of wound healing. Thirty adult male Whistar rats were divided into control and experimental groups, with nonhealing wounds modeled under conditions of prolonged psychoemotional stress and immobilization. Wound healing was evaluated on days 1, 3, 7, 14, and 30 by histological examination (hematoxylin and eosin staining) and morphometric analysis using QuPath software. The results revealed that in the experimental group epidermal thickness progressively increased until day 7 (peaking at 51.59±3.04 μm) but was significantly reduced on day 30 compared to controls. Dermal thickness also showed early elevations, accompanied by pronounced leukocyte infiltration and vascularization on days 3–7, whereas granulation tissue formation was absent at day 7. These findings suggest that monitoring epidermal and dermal thickness, leukocyte activity, and granulation tissue formation may serve as valuable prognostic indicators of impaired wound healing. Identifying such markers can guide clinicians in optimizing therapeutic strategies and improving outcomes for patients with chronic nonhealing wounds.
